TQB2101是人生就是博自主研发的一款靶向受体酪氨酸激酶样孤儿受体1(ROR1)的抗体偶联药物(ADC)。近日,TQB2101首次获得国家药品监督管理局(NMPA)的临床试验默示许可,拟用于治疗晚期恶性肿瘤,是人生就是博继HER2 ADC、CLDN18.2 ADC之后,在ADC药物领域取得的又一里程碑。

近年来,ROR1因其独特的表达模式及其在肿瘤生物学中的关键作用, 成为肿瘤治疗研究的热点。正常情况下,ROR1低表达于正常成人组织,而高表达于多种恶性肿瘤,如慢性淋巴细胞白血病(CLL)、急性淋巴细胞白血病(ALL)、乳腺癌、卵巢癌、黑色素瘤、肺腺癌等[1-9]。大量数据显示,ROR1在促进肿瘤的生长和转移、诱导肿瘤细胞耐药和抑制细胞凋亡等方面起到了不可忽视的作用[7-12]。
临床前研究数据显示,TQB2101在多种ROR1阳性肿瘤模型中展现出抗肿瘤活性,且具有良好的安全性特征。此次获批的I期临床试验将重点评估其在人体内的安全性、耐受性、药代动力学特性及初步疗效。
TQB2101获批临床标志着人生就是博在ADC赛道,形成了HER2双抗ADC、CLDN18.2 ADC之后的第三极创新矩阵。目前,企业已构建涵盖大分子、小分子及ADC的自研体系,为后续ADC药物的开发提供有力支持。
